چکیده
Purpose: To determine the expression of circRTN4 in gastric cancer (GC) and its role tumor progression.
Methods: GEO dataset GSE93541 was analyzed using GEO2R. Starbase website used to predict combination miRNA circRTN4. The relationship between prognosis Kaplan-Meier Plotter database, while levels circRTN4, miR-424-5p, LATS2 were assessed by quantitative real time-polymerase chain reaction (qRT-PCR). CCK8, EDU, Transwell, Western blot assess GC proliferation, migration, invasion, stemness. Lastly, co-transfection miR-424-5p or si-LAST2 reversely demonstrate regulatory effect on progression cells. Significantly downregulated screened, combined downstream gene predicted Starbase.
Results: average relative mRNA tissues significantly lower than adjacent tissues. MiR-424-5p highly expressed tissues, decreased (p < 0.05). Low associated with a low survival rate patients. pLCDH- inhibited proliferation cells Overexpression migration invasion cells, pLCDH-circRTN4 reduced ability stem expressions MMP2 OCT4.
Conclusion: Expression decreases GC, contributes development this disease increasing binding miR-424-5p. This suggests that may serve as promising prognostic marker well potential therapeutic target for

miR 1296-5p Inhibits the Migration and Invasion of Gastric Cancer Cells by Repressing ERBB2 Expression
The metastasis of gastric cancer, one of the most common tumors, has a molecular mechanism that is still largely unclear. Here we investigated the role of possible tumor-suppressor miR-1296-5p in the cell migration and invasion of ERBB2-positive gastric cancer. It found that miR-1296-5p was significantly down-regulated in gastric cancer tissues. Moreover, it was down-regulated in lymph node met...
متن کاملmiR-424-5p promotes proliferation of gastric cancer by targeting Smad3 through TGF-β signaling pathway
MiRNAs have been reported to regulate gene expression and be associated with cancer progression. Recently, miR-424-5p was reported to play important role in a variety of tumors. However, the role and molecular mechanisms of miR-424-5p in GC (gastric cancer) remains largely unknown. In this study, we aimed to explore the role of miR-424-5p in GC. QRT-PCR was used to determine the expression leve...
متن کاملSaffron Aqueous Extract Inhibits the Chemically-induced Gastric Cancer Progression in the Wistar Albino Rat
Objective(s): Gastric cancer is the first and second leading cause of cancer related death in Iranian men and women, respectively. Gastric cancer management is based on the surgery, radiotherapy and chemotherapy. In the present study, for the first time, the beneficial effect of saffron (Crocus sativus L.) aqueous extract (SAE) on the 1-Methyl-3-nitro-1-nitrosoguanidine (MNNG)-induced gastric c...
متن کاملDecreased miR-155-5p, miR-15a, and miR-186 Expression in Gastric Cancer Is Associated with Advanced Tumor Grade and Metastasis
Background: Gastric cancer (GC) is one of the most prevalent cancers with a high rate of mortality in the world. In recent years, microRNAs (miRNAs) have been proposed to be involved in GC development. In this study, we aimed at investigating differential expression level of miR-155-5p, miR-15a, miR-15b, and miR-186 in GC. Methods: For this research, we used qPCR to investigate miR-15b, miR-155...
متن کاملA genome-wide microRNA profiling indicates miR-424-5p and miR-503-5p as regulators of ALK expression in neuroblastoma
The discovery of missense mutations of ALK gene identified this receptor tyrosine kinase as a therapeutic target in neuroblastoma (NB). Moreover, a high level of ALK protein has been associated with metastatic NB cases and with a worse prognosis, suggesting that also ALK overexpression is involved in NB tumorigenesis. Since miRNAs play key roles in the regulation of gene expression we aimed at ...
